Constructs are expressed from the Adh1 promoter or the Sec2 promoter as indicated. (B) Serial dilutions of the overnight cultures from indicated strains were plated on YP Galactose plates to induce protein expression under the Gal1 promoter and grown for 3 days at different temperatures as shown. Constructs are in either a sec2 Δ background or are co-expressed with endogenous Sec2 as indicated. Strains exhibiting mild growth defects are framed. (C) Serial dilutions of the overnight cultures from indicated strains were plated on YPD plates and grown for 2 days at different temperatures as shown. Constructs are in a vps4Δ or endogenous VPS4 backgrounds.
