Doxorubicin recruits IGF2BP and increases NLRP3 expression by upregulating TINCR, which in turn activates caspase1, cleaves GMDSD-N, and releases IL-1β, IL-18. Doxorubicin activates Bnip3 in mitochondria, which activates caspase3 and causes GSDME-dependent pyroptosis.
