Genome editing of somatic cells using JoinTRV

Plants were agroinoculated with an Agrobacterium strain that simultaneously hosts pLX-TRV1 and pLX-TRV2::sgPDS3-tFT, a pLX-TRV2 derivative with a sgRNA targeting two N. benthamiana PDS homeologs.

(A) Phenotypes of agroinoculated wild-type and Cas9 plants (∼1 month post inoculation).

(B) Severe photobleaching phenotype in Cas9 mature plants (∼2.5 months post inoculation).

(C) Chromatogram deconvolution of edited and control samples from agroinoculated Cas9 plants; note the high percentage of aberrant sequence downstream the predicted Cas9 cleavage site (blue dashed line).
