Left frame summarizes the transfection and monitoring of GFPO fluorescence procedure. The right frame shows that A3A, in absence of inhibitor, corrects Gfp to express functional GFP. The Cas9n is directed to the Gfp 5’-TCA site-of-interest using a complementary gRNA and opens an R-loop that A3A (here unlinked to Cas9n) edits to 5’-TTA to restore GFP functionality. PS-TTFdZ-hairpin inhibits this DNA-editing by A3A. Left panel schematic was designed using BioRender.
