Figure describes eleven methods for mediation analysis that can be applied to high-dimensional DNA methylation data, each of which is described in greater detail in the Methods section. Seven of these methods are included in the simulation study and nine in the observed DNAm data analysis with MESA. Group 1 methods fit the outcome model explicitly using penalized regression or Bayesian regresion; Group 2 methods obtain the global mediation effect without fitting the original outcome model explicitly; and Group 3 methods measure mediation through latent variables.
