The N and C termini of TERT are necessary for telomerase activity. (A) Telomerase activity assays were performed with equal amounts (3 μl) of rabbit reticulocyte lysate reaction mixtures coexpressing TERT or the TERT truncation indicated and WT telomerase RNA. Activity was monitored by the incorporation of radiolabeled dGTP and unlabeled dTTP to elongate the primer (TG)8TTG. (B) Equal amounts (0.5 μl) of each [35S]methionine-labeled TERT and telomerase RNA coexpression reaction were analyzed by electrophoresis on an SDS–15% PAGE gel and subsequent autoradiography.
