Enhancement of the endosomal escape of Cy5-siRNA in RPE cells by STR-CH2R4H2C-LP-siRNA Lipoplex. Naked Cy5-siRNA, DOTAP0.250-LP-siRNA lipoplexes, 2.0 mol% STR-CH2R4H2C-neutral-LP-siRNA lipoplexes, and 2.0 mol% STR-CH2R4H2C-DOTAP0.250-LP-siRNA lipoplexes (Cy5-siRNA, 0.079 µM) were transfected into RPE cells (3.0 × 10⁵ cells) in serum-free DMEM for 4 h. Cells were washed in PBS and then incubated with 75 nM LysoTracker^TM Red DND-99 for 1 h followed by sealing in SlowFade^® containing 1% Hoechst 33342. Cy5-siRNA localization in the cytoplasm was observed using confocal laser scanning microscope. Magnification: 800×; scale bar: 50 µm; yellow: Cy5-siRNA; blue: nucleus; red: endosome.
