The CD47-SIRPα signal axis. The interaction between CD47 and SIRPα recruits SHP-1/2, leading to the phosphorylation of two tyrosine residues of ITIMs in the intracellular structural domain of SIRPα. Phosphorylation of SHP-1/2 prevents the accumulation of myosin-IIA, which forms phagocytic synapses, resulting in sending a “don’t eat me” signal. Blocking the CD47-SIRPa axis could restore phagocytosis.
