(A) Schematic methodology: SCN explants from a Per2Luc mouse producing the PERIOD2::LUCIFERASE fusion protein as a reporter of PER2 expression were cultured on organotypic membranes inside Petri dishes from which bioluminescence was recorded using photomultiplier tubes (PMT). (B) PER2 expression single traces from adult SCN explants in vehicle (VEH; n=11) and three representative ammonia concentrations (n=6-10) along six days of recording. (C) Mean±SEM group trace comparisons for VEH (continuous line) and ammonia 5 mM (dashed line). (D) Ammonia induced a reduction in amplitude measured at the second day of recording of ~80% at and above ammonia 0.1 mM (*p=0.002) while period (E) was increased also at and above ammonia 0.1 mM (*p=0.003).
