Point mutations at positions T-14 and T-15 in the Δ42 allele have the greatest effect on growth. Cells were grown overnight in YEPD followed by plating fivefold dilutions on 5-FOA, −Trp. 1X equals an optical density at 600 nm of 0.13. Plates were incubated at 30°C for 2 days.
