Steady-state levels of TFIID and SAGA subunits following a temperature shift. Wild-type and mutant strains were grown at 24°C in YPAD to an OD₆₀₀ of approximately 0.4, and after removal of an aliquot for the t = 0 time point, they were transferred to a 37°C shaking water bath. Aliquots of culture were removed after 1, 2, and 4 h at the nonpermissive temperature. Extracts prepared from these cells (20 μg of protein) were fractionated by SDS-polyacrylamide gel electrophoresis and transferred to nitrocellulose, and the specific proteins were detected by immunoblotting.
