Flow cytometric analysis of apoptosis in thymocytes after various treatments. Ovariectomized mice (100 days old) were treated for 7 days with DMSO (Left), 5 mg/kg per day of E2 (Center), or 80 mg/kg per day of genistein (Right); thymocytes then were isolated and stained for annexin-V labeled with FITC and PI. Normal cells are seen in the lower left quadrant of each panel. Thymocytes showing annexin-V staining, an early apoptotic event, are seen in the lower right quadrant of each panel. (Upper) Cells in later stages of cell death, when they are permeable to PI as a consequence of plasma membrane ruptures. As expected, essentially all cells permeable to PI also showed annexin staining (upper right quadrants), so few cells stained for PI but not annexin (upper left quadrants).
