CD28 costimulation enhances nuclear factor of activated T cells (NFAT) DNA-binding activity in nuclear extracts. Nuclear extracts from stimulated T lymphocytes were made as in Fig. 10. Nuclear extract (7.5 mg) was assessed for NFAT DNA-binding activity in a gel shift assay by using an IL-2 promoter probe [–255 to –285; see Shaw, J. P., Utz, P. J., Durand, D. B., Toole, J. J., Emmel, E. A. & Crabtree, G. R. (1988) Science 241, 202–205]. Nuclear extract from Jurkat T cells stimulated with ionomycin and PMA was used as a positive control for NFAT DNA-binding activity.
