The inhibitory effect of the Gq/phospholipase C b (PLCb) signaling pathway on Wnt/b-catenin cascade is not mediated by PKC but mimicked by thapsigargin. HEK293 cells stably expressing human Frizzled-1 were transfected with control, Wnt-3a, and Q209L-Gaq expression plasmids. PLCb inhibitor U-73122 (5 mM), PKC inhibitors BIM-I (100 nM), and calphostin C (100 nM) were added on transfection, and thapsigargin (50 nM) was added 24 h after transfection. Relative TCF/LEF-1-induced transcriptional activity was assayed 48 h after transfection.
