(A) Sagittal brain slides from 12-month-old APP23/Ar+/- animals were immunostained with anti-Ab and CD11 and showed colocalization of activated microglia and plaques. Microglia were isolated from WT and Ar+/- mice at postnatal 0-3 days and incubated with fluorescent-labeled Ab (100 nM) for 24 h with 24-h pretreatment of 17b-estradiol (100 nM) in combination with ICI 182,789 or vehicle. (B Upper) A decrease in Ab uptake was observed in the Ar+/--derived microglia compared with the microglia from WT mice at 6- and 24-h incubation. (B Lower) Estrogen treatment reversed the impairment of Ab uptake in Ar+/--derived microglia with partial dependency on estrogen receptors.
