Leaves of the transgenic flax line Hosh::M, carrying the M resistance gene, were infiltrated with A. tumefacienscultures containing T-DNA plasmids encoding AvrM-A, -B, -C,¬ or -D either as the full length gene with the signal peptide (SP), or as a truncated gene lacking the signal peptide (δSP)¬. Images prepared 9 days after infiltration.
- Supplemental Figure 2 - Supplemental Figure 2. Activity of AvrM and AvrP4 Signal Peptides in Yeast. DBYα2445 yeast cells, deficient in invertase activity, were transformed with the vector pSMASH V2 containing either no insert (empty vector), a single inserted ATG codon (methionine), the yeast suc2 invertase SP, the first 32 amino acids of either AvrM or AvrP4, or the 44 amino acid leader sequence of bean phytohemagglutinin (PHA44). Transformed cells were grown overnight in minimal media with glucose and then resuspended in water at an OD600 of 0.01. Ten μl aliquots of undiluted culture (left column) and ½, ¼, 1/16, 1/32, and 1/64 dilutions were inoculated onto YP sucrose media and photographed after 2 days growth at 30°C.
