Western 2D gel analysis and protein identification from AMD Bruch’s membrane/RPE/choroid tissue. About 200 µg protein from Bruch’s membrane/RPE/choroid from an 84-year-old female AMD donor was isoelectric focused on an 18-cm immobilized pH gradient strip (88, 402 V) then separated by SDS/PAGE on a large gel (23.5´18 ´0.1 cm). After electrophoresis, protein was partially transferred to poly(vinylidene difluoride) membrane. (A) The gel stained with collodial Coomassie blue and (B) the membrane probed by Western analysis with rabbit polyconal anti-CEP antibody. Immunoreactive spots were excised from the gel and digested with trypsin, and the indicated proteins were identified by peptide mass mapping using matrix-assisted laser desorption ionization-time-of-flight MS as described in Methods.
