Both gold-UbcH5C and nanogold-Ubq are competent and specific components of the ubiquitination reaction, as visualized by EM (Fig. 3), and Western blotting against Ubq. BARD1/BRCA1/gold-UbcH5C (Body:gold-UbcH5C) efficiently activates polymerization of nanogold-Ubq, and this activity is specific as removal of gold-UbcH5C (- UbcH5C), E1 (- E1), and ATP (- ATP) prevents ubiquitin conjugation. Gold-UbcH5C alone (UbcH5C) is not sufficient to polymerize nanogold-Ubq.
