Levels of EKLF protein are similar in whole-cell lysates from G1E and G1E-ER-GATA-1 cells. Cells were treated with 1 m M tamoxifen for 48 h. Whole-cell lysates were isolated, resolved on an SDS/10% polyacrylamide gel, and analyzed by Western blotting with the anti-EKLF 4B9 mouse monoclonal antibody (from James Bieker, Mt. Sinai Medical Center, New York) as described [Johnson, K. D., Grass, J. D., Boyer, M. E., Kiekhaefer, C. M., Blobel, G. A., Weiss, M. J. & Bresnick, E. H. (2002) Proc. Natl. Acad. Sci. USA 99, 11760–11765]. The arrows depict endogenous EKLF isoforms. Each lane represents lysate from 500,000 cells. The result is representative of two independent blots with different lysates.
