Fig. 6. TASK-3 channels are appropriately expressed and targeted to membranes of Gaq*-expressing HEK293 cells. HEK293 cells were transfected with hemagglutinin (HA)-tagged TASK-3 and constitutively-active Gaq (Gaq*) as indicated. Membrane proteins were biotinylated, and crude membrane preparations were precipitated by using streptavidin-linked agarose beads. Western blots detecting (HA)TASK-3 immunoreactivity in the membrane preparation (Left) and in the avidin-IP (Right) reveal that expression and surface targeting of TASK-3 were unaffected by the cotransfected Gaq*. Control streptavidin precipitates obtained from non-biotinylated cells were devoid of HA immunoreactivity (Right, lane 1). Data are representative of two independent experiments.







Fig. 7. Activated Gaq and Ga11 subunits coassociate with TASK channels. HEK293 cells were transfected with HA-tagged TASK-1 and constitutively-active Gaq and Ga11, as indicated (Gaq* & Ga11*); control cells were transfected with GFP alone (q). We retrieved Gaq* and Ga11* in HA-immunoprecipitates from cells cotransfected with HA-tagged TASK-1; note that immunoprecipitation of Ga subunits required expression of the HA-tagged channel.
