Movie 1. Video recording of a pulling experiment, which leads to detachment of a C. crescentus cell from a flexible micropipette. A single cell is trapped, held onto, and then pulled by a suction micropipette. Initially phase-contrast images show the relative position of the two micropipettes. The recording is then switched to florescence imaging, which shows the position of the cell (pointed to by the red arrow). The cell was labeled by Nano-Orange (Molecular Probes). The pulling of the cell causes bending of the capillary until the cell suddenly detaches. The detachment force is determined by the extent of bending as the cell detaches.
