GFP shuffling. Reassembly reactions were done with three different initial fragment concentrations and three different digestion conditions. Equivalent volumes of the nine total reactions were analyzed by gel electrophoresis. An ≈750-bp band is apparent at all conditions. Heavily digested fragments [average fragment size (AFS) 45 bp, minimum fragment size (MFS) 25 bp, lanes C, F, and I] have less apparent peaks and larger smear, including an ≈1.3-kb multimer. Peaks are less distinct at higher concentrations (120 ng/µl) (G, H, I), and there is more of a smear compared to lower concentrations (8 ng/µl).
