RT-PCR detection of rpmH-rnpA mRNA by using total RNA isolated from T. thermophilus HB8; mRNA portions covering the following amino acids of C51 could be amplified (see also Fig. 1): amino acids 2–51 (primers A'/A, lane 1), 37–75 (primers B'/B, lane 2), 71–107 (primers C'/C, lane 3), 2–107 (primers A'/C, lane 4), and 2 to the stop codon (primers A'/D, lane 5). Lanes 6–10: as lanes 1–5, but omission of reverse transcriptase. M, double-stranded DNA marker (Life Technologies, 100-bp DNA ladder) with sizes indicated on the left. A photograph negative of an ethidium bromide-stained 2.5% agarose gel is shown. This experiment demonstrates the presence of full-length rnpA mRNA in T. thermophilus.
