Oligonucleotide synthesis by mutationally altered primases on M13 DNA. Oligonucleotide synthesis assays were performed on M13 DNA with either ATP and [a-32P]CTP or GTP and [a-32P]CTP as indicated. After synthesis, products were separated on a 25% denaturing PAGE gel and autoradiographed. Mutationally altered primases employed in each reaction and the oligonucleotide products synthesized are identified. For comparison, the oligonucleotide products synthesized by wild-type T7 primase are also illustrated.
