Single-strand conformation polymorphism detection of an alteration in the genome of the mud⁴ line. A nondenaturing gel was loaded with the indicated restriction digests of genomic DNA extracted either from the three alleles of mud that were derived by chemical mutagenesis from the Berlin strain or from Vam, a stock also derived from Berlin but bearing a mutation in an unrelated gene. After overnight electrophoresis, the gel was transferred to a nylon membrane and hybridized with an XbaI fragment of cos80G4 that covered coordinates 54-62 of the walk shown in Fig. 1. Arrowheads point to fragments whose mobility is distinctive in mud⁴ DNA.
