- Supplemental Figure 1 - The tagged Mvb12 derivatives Mvb12-HA, Mvb12-GFP and HA-Mvb12 are functional. mvb12· cells expressing GFP-CPS were transformed with empty vector or a plasmid containing each of the MVB12 fusions and analyzed by fluorescence microscopy.
- Supplemental Figure 2 - Sna3-GFP and Ste3-GFP trafficking phenotype in cells deleted for VPS23 or MVB12. The indicated strains expressing either Sna3-GFP or Ste3-GFP were analyzed by fluorescence microscopy. The effect of ESCRT-II overexpression on the trafficking phenotype was observed using pMB175, a high-copy plasmid expressing all three subunits of ESCRT-II (2μ ESCRT-II). Arrows indicate class E compartments.
