Genomic DNA was amplified to obtain part of class II a1 domain sequences from the first nurse shark family. The conditions were 94ºC for 4 min, followed by 40 cycles of 94ºC for 1 min, 63ºC for 1 min, 72ºC for 1 min, and a final extension at 72ºC for 15 min, with the previously described primer set, primer 74 and primer 78 [Kasahara, M., McKinney, E. C., Flajnik, M. F. & Ishibashi, T. (1993) Eur. J. Immunol. 23, 2160-2165]. The 275 base pair products were then cloned into pCR2.1 TA-cloning vector (Invitrogen) and sequenced. Gici-DAA*01-06, Gici-DBA*01-05 were reported previously [see ref. above]. We added new alleles found in this family. The paternal alleles identified in this study were: DAA*02, sib 7,12,16; DAA*04, sib 1,2,5,6,9,11,14,15,17; DAA*05, sib 8; DAA*07, sib 3; DBA*06, sib 13.
