Accuracy of molecular beacon-based genotyping assays. This figure illustrates the ability of molecular beacon assays to unambiguously discriminate for CCR5-627C and CCR5-627T whenever CCR5-630 is a CCR5-630C. Data from this assay were not used when the CCR5-630 position is CCR5-630T. The CCR5-630 SNP was determined by a PCR-RFLP genotyping assay. Fluorescein fluorescence at the 35th cycle is plotted against TET fluorescence. The data shown are from the WHMC cohort. Each sample (circle) falls into one of four easily distinguishable categories: (i) high fluorescein fluorescence and low TET fluorescence (green); (ii) high TET fluorescence and low fluorescein fluorescence (red); (iii) high fluorescein fluorescence and high TET fluorescence (orange); and (iv) low fluorescein fluorescence and low TET fluorescence (negative controls, blue). The entire fluorescence vs. cycle profiles were analyzed for the samples that produced little fluorescence signal at the 35th cycle.
