Fig. 8. (A) Quantitation of human tau levels in transgenic lines. Relative levels of human tau expressed in transgenic lines were determined by quantitative immunoblot analysis of whole animal lysates. T46 monoclonal antibody was used to detect tau and monoclonal antibody E7 was used to detect endogenous Caenorhabditis elegans β-tubulin. The ratio of tau to β-tubulin is given below each lane. (B) Titration of transgene copy number. Extrachromosomal transgenic lines with varying tau expression were generated by microinjection by using increasing concentrations of tau transgene DNA. The aex-3::tau transgene was injected at concentrations ranging from 0 to 120 ng/ml; total injected DNA remained fixed at 125 mg/m l, with pBluescript making up the difference and 5 ng/ml of myo-2::gfp transgene as marker. The severity of the uncoordinated locomotion (Unc) phenotype was measured using the liquid thrashing assay for 10 animals from each line. Error bars are the SEM.
