- Figure S1. One month old Tax+ mice show normal osteoclasts both in vivo and in vitro (PDF, 1.96 MB) - A. Osteoclasts cultured from bone marrow macrophages were evaluated for production of the osteoclast specific marker TRAP. No difference in TRAP production was observed between wild type and Tax+ osteoclasts from one month old pre-leukemic mice (p>0.05). All data are depicted as the mean+/- s.e.m. B. RT-PCR of bone marrow osteoclasts from Tax+ mice failed to express granzyme B (the promoter driving Tax+ in this animal model), whereas purified bone marrow derived murine NK cells expressed granzyme B. C. Histomorphometric analysis on TRAP stained decalcified tibias reveals normal osteoclast numbers per osteoblast surface (OB) in Tax+ mice at one month of age compared to wild type littermates. All data are depicted as the mean+/- s.e.m. (p>0.05).

Isolated Tax tumor cells express RANK but not OPG (PDF, 82.8 KB) - RT-PCR for RANK and OPG was performed using single cell suspensions of two Tax+ tumors from two separate mice which were sorted by FACS according to FcR II/III (CD16/32) FITC negative (N), low and high (H) intensity. GAPDH was used as a loading control.
