Ultrastructual analysis of ama1D mutants. Wild-type (A and B) and ama1D mutant (C and D) cultures were fixed 24 h following shift to sporulation medium as described [Neiman, A. M. (1998) J. Cell Biol. 140, 29–37] to enhance visualization of the spore wall layers and membranes. The wild-type spore wall contains the inner spore wall (open arrow), two chitosan/chitin layers that appear as a single layer (Ch), and the electron-dense dityrosine outer layer (filled arrow). The putative prospore wall in the mutant (large arrows) and dark staining cytoplasmic vesicles (small arrows) are indicated in the ama1D mutant. N, nucleus.
