KIR2DL2+ T cells were coincubated with either SEB-coated 721.221 cells or with 721.221/HLA-Cw3 cells. T cells were incubated for 30 minutes at 4°C with the anti-KIR2DS2/DL2/3 mAb CH-L, washed twice with PBS, and then resuspended in complete media. 50,000 T cells were co-cultured with 25,000 SEB-coated 721.221/HLA-Cw3 or 721.221 cells for 48 hours. IFN-
secreted into the supernatant was quantified by ELISA. Results are shown as mean ± SD of triplicate cultures. CH-L at the concentration used did not block KIR2DL2 function and was therefore used with the same experimental conditions in the confocal imaging experiments.
