The single (R243Q, D245Q) and double mutations (R243Q+D245Q) that modified the V_m gating of RCx43 junctions did not have any effect on V_j gating. Sample records of junctional currents (I_j) evoked by V_j pulses (±100 mV in 20-mV increments, 5 s) in wild-type and mutated homomeric junctions. The comparison among the four types of junctions shows that their junctional currents decreased to progressively lower values and faster at larger V_j’s with similar voltage sensitivity and kinetics. Positive and negative V_jpolarities are defined as relative depolarization and hyperpolarization of the cell where the pulses were applied and the corresponding I_j’s plotted as upward and downward respectively.
