The single (R243Q, D245Q) and double mutations (R243Q+D245Q) that modified the Vm gating of RCx43 junctions did not have any effect on Vj gating. Sample records of junctional currents (Ij) evoked by Vj pulses (±100 mV in 20-mV increments, 5 s) in wild-type and mutated homomeric junctions. The comparison among the four types of junctions shows that their junctional currents decreased to progressively lower values and faster at larger Vjs with similar voltage sensitivity and kinetics. Positive and negative Vj polarities are defined as relative depolarization and hyperpolarization of the cell where the pulses were applied and the corresponding Ijs plotted as upward and downward respectively.
