- Supplemental 1 - The cell-permeable peptide AP-NES does not affect the cellular distribution of HuR in undifferentiated C2C12 cells. Exponentially growing C2C12 cells untreated (No peptide) or treated with the AP-NES cell-permeable peptide for 24 hours were fixed and the cellular localization of HuR was determined by immunofluorescence using the monoclonal anti-HuR antibody. DAPI staining was used to show nuclear integrity. DAPI, HuR, and a merged image of a single representative field of view are shown for each treatment.
- Supplemental 2 - The cell permeable peptide AP-HNS enhances myogenesis on Day 2 and Day 4 of differentiation. Exponentially growing C2C12 cells were treated with the indicated AP-conjugated peptides, and the degree of differentiation after treatment was observed by phase contrast microscopy on both Day 2 and Day 4 of differentiation. A single representative field of view for each peptide treatment is shown for the indicated days of differentiation.
