Fluorescent titration of Fluo-4 with Ca²⁺. Shown is the percent of maximal fluorescence versus the calculated [Ca²⁺] (μM). The titration produced the expected noncooperative binding curve and a Fluo-4 K_d for Ca²⁺ of 450 ± 50 nM was determined, a value close to the literature K_d of 365 nM. The agreement between the K_d of Fluo-4 calculated from this experiment with its literature value confirms that the Ca²⁺/EGTA buffer is accurately calibrated to give the proper [Ca²⁺].
