- Supplemental Figure 1 - PKA only phosphorylates β4 S1364 in vitro. COS-7 cells were transiently transfected with either IL2R-β4WT (panel A), IL2R- β4S1360A (panel B), IL2R-β4S1364A (panel C) cDNA constructs or empty vector (panel D). The cells were lysed in mPER buffer and IPs were done using the β4 mAb 450-11A. The isolated β4 chimeric proteins were phosphorylated in vitro by PKA and phosphopeptide mapping was performed. Panel E is a schematic diagram of the results shown in panels A-D. Grey ovals - unidentified phosphopeptides.
