(A) Wild-type (wt) and esp1-1 cells expressing wild-type Bub2 (strains ySP41 and ySP278), Bub2-HA6 (strains ySP3867 and ySP3964), or Bub2-myc9 (strains ySP710 and ySP3786) were grown at 25deg;C, arrested in G1 with α factor, and released at 37°C, which is the restrictive temperature for esp1-1. Cells were collected at the indicated time points for FACS analysis of DNA contents. (B) Examples of asymmetric localization of Bub2-HA6 and symmetric localization of Bub2-myc9 at SPBs in esp1-1 cells after immunostaining with anti-HA and anti-myc antibodies, respectively.
