(A) GFP-tagged MVB cargoes Sna3 (biosynthetic cargo) and Ste3 (endocytic cargo) were visualized by fluorescent and brightfield microscopy in living cells corresponding to wild type, vta1Δ, and the known class E vps mutant vps4Δ in three distinct genetic backgrounds. Arrowheads highlight the class E compartment in vps4Δ cells. (B) ESCRT-III recycling was addressed in wild-type and vta1&Delta cells representing the BY4742 and SF838-9D backgrounds by performing differential centrifugation on extracts generated from the indicated strain. The resulting soluble (S) and pellet (P) fractions were probed with anti-Snf7 antibody.
