Figure S3. MyD88 is required for IL-6 secretion and CD40 and CD69 up-regulation. (a) IL-6 secretion was measured from wild-type and Myd88^-/- splenocytes stimulated for 12 h with CpG. As indicated, cells were pretreated with 10 µM chloroquine for 15 min. ND indicates that no IL-6 secretion was detected. (b) CD40 and CD69 expression level was measured as described in Materials and methods on the surface of gated B cells from the splenocytes suspension. Grey histograms represent untreated cells, black histograms represents CpG-stimulated cells, and discontinuous histograms represent CpG-treated cells that had been pretreated for 15 min with 10 µM chloroquine.
