Figure S1. Testing P-SCG10 antibody specificity for immunostaining. To test the specificity of the P-SCG10 antibody, COS-7 cells were transfected with CFP-SCG10-AA or -WT + GFP-MLK3 to activate JNK. Following 24 h of expression, cells were fixed and stained with P-SCG10 antibody (1:1,000). P-SCG10 immunoreactivity was detected using anti-rabbit Alexa-546 antibodies (1:500). CFP-SCG10 expression is shown in the lower panels and P-SCG10 immunoreactivity in corresponding cells in the upper panels. Equivalent acquisition parameters were used. P-SCG10 did not significantly cross-react with CFP-SCG10-AA.
