Figure S1. Cdc5 protein levels and associated kinase activity are markedly delayed upon CLA4t overexpression. Wild-type (wt; ySP939) and 4X GAL1-CLA4t (ySP3346) cells expressing HA-tagged Cdc5 (Cdc5-HA3) were grown in YEPR at 25°C, arrested in G1 by α factor, and released in YEPRG at 25°C (time 0). At the indicated time points, cells were collected for FACS analysis of DNA contents (histograms) and protein extracts for immunoblotting with anti-HA antibodies and Cdc5 kinase assays using casein as substrate. Swi6 was used as loading control.
