Figure S3. RNAi knockdown of gw mRNA phenocopies the gw1 mutation. Each image represents a maximum projection of 80 1-µm sections. Embryos expressing nuclear GFP were injected with 500 pl of a 1-µg/µl gw 3′ untranslated region double-stranded RNA and were imaged at 2 h 10 min AED. (A) In embryos injected with gw double-stranded RNA (arrowheads) at 30 min AED, the majority of nuclei are lost. (B) RNAi knockdown of gw 1 h AED leads to the presence of expanded nuclei in the interior and far fewer nuclei at the cortex. (C) The effectiveness of the gw dsRNA was confirmed by incubating it with cultured S2 cells. There is a progressive knockdown of GW protein levels relative to tubulin over 3 d.
