Figure S3. Effects of BAPTA and thapsigargin on cytochrome c release and cell death. (A) Cytochrome c distribution in Jurkat cells preloaded with 500 nM BAPTA-AM and treated with 1 ng/ml FasL for 0, 12, or 24 h. This concentration of BAPTA was determined in separate experiments to be the highest dose that was not toxic over the time course of the experiment. (B) Caspase (DEVDase) activity in control and BAPTA-loaded Jurkat cells in response to FasL. (C) Cell death in control and BAPTA-loaded Jurkat cells treated with FasL. (D) Cytochrome c distribution in Jurkat cells treated with 0, 0.5, or 2 µM thapsigargin (TG) for 24 h. Caspase (DEVDase) activity (E) and cell death (F) in Jurkat cells treated with TG for 24 h.
