(A) HeLa cells were synchronized and transfected as for Fig. 3 with control (siCTR) or a second siRNA duplex targeting Emi1 (siEmi1_2), and samples were collected 0, 12, and 20 h after aphidicolin release for flow cytometry analysis. Representative of two experiments. (B) Hela cells were transfected as for A, and mitotic entry was analyzed by time-lapse DIC microscopy. The cumulative number of cells entering mitosis (NEBD) is plotted over time. Note that the siRNA oligonucleotide used in the right panels was siEmi1_2, not siEmi1_1.
