Fig. 5. Cells (11,1) were transiently transfected with wild-type human Tyk2 or the indicated mutants (E782K, R856G, and K930R). Forty-eight hours after transfection, 1-1.5 mg of cell lysates were immunoprecipitated with R5-9 Ab and probed with phosphotyrosine-specific Abs (4G10; Upper). To demonstrate equal protein expression and protein loading, the same membrane was reprobed with T10-2 monoclonal anti-human Tyk2 Ab. K930R, a kinase-dead mutant described in ref. 1, was used as negative control.
