Figure S2. L-22 does not act on naive or differentiated T cells. (A) DO11 cells were differentiated to Th1, Th2, and Th17 for 7 d with conditions as described in Fig. 2 A. Expression of IL-22R1 transcript was examined in naive and differentiated DO11 T cells by quantitative PCR. Skin RNA was used as a positive control. (B) Naive DO11 T cells were activated in the presence of IL-22, irradiated splenocytes, and OVAp. On day 4, cells were assayed for proliferation by [3H]thymidine incorporation. To examine the effect of IL-22 on differentiated T cells generated as described in Fig. 2 B, Th1 (C), Th2 (D), and Th17 (E) cells generated from a primary stimulation were restimulated in the presence of IL-22. IFN-γ, IL-4, and IL-17A were measured on day 4.
