Two siRNAs targeting JAM-C (JAM-C-siRNA-1 and JAM-C-siRNA-2), and respective control siRNAs with a single mutation (Mut-siRNA-1 and Mut-siRNA-2), and a nontargeting siRNA (Con-siRNA) and a siRNA against an irrelevant protein (cyclophilin-siRNA) were engaged. (A) HDMECs that were transfected with different siRNAs, as indicated, were incubated for 1 h without (-) or with VEGF (50 ng/ml) or histamine (50 µM). Permeability is expressed as the percentage of control, defined as permeability of the Mut-siRNA-1-transfected HDMECs in the absence of any stimulus.
