Figure S3. Characterization of FSAP-mediated cleavage of PDGF-BB. Western blot analysis of PDGF-BB (1 µg/ml) incubated with (A) FSAP (12 µg/ml) in the presence of heparin (10 µg/ml) for the indicated times at 37°C (B) increasing concentration of FSAP (0.05-12 µg/ml) in the presence of heparin (10 µg/ml) for 1 h at 37°C. In the final lane the highest concentration of FSAP (12 µg/ml) was added in the presence of aprotinin (10 µg/ml). Western blots were performed with an anti-PDGF-BB antibody under reducing and nonreducing conditions. This preparation of PDGF-BB was obtained from Immunotools and displayed a partial double-band character under reducing conditions.
