Bone marrow samples from animals of the indicated genotypes were obtained from both femora. Aliquots of single-cell suspensions were counted in a hemocytometer using trypan blue to determine the overall cellularity. Total numbers of the different hemopoietic subpopulations were determined by staining aliquots with fluorescence-conjugated antibodies specific for the myeloid markers Mac-1 and Gr-1, the erythroid cell marker Ter-119, the T cell markers CD4 and CD8, or the B cell markers B220, sIgM, and CD43 followed by flow cytometric analysis. Means ± SEM of 5–10 animals per genotype are given. DKO, double knockout. |
