Figure S2. Phenotype, longevity, and TCR avidity are similar in memory cells generated in wild-type or CD40KO mice. Wild-type (C57BL/6) or CD40KO mice were primed with H19-Env peptide-pulsed wild-type DCs s.c. (A) After 5 wk, the mice were boosted with peptide in CFA and cells in the draining lymph node were analyzed on day 5. The dot plots show the percentage of tetramer+ CD4 T cells that are CD62Llo (two left-hand plots) or CD44hi (two right-hand plots). Below are the histograms, gated on the rectangle dot plot gate showing intensity of tetramer, CD62L, or CD44 staining in wild-type (bold lines) or CD40KO mice (dashed lines). (B) After 5 wk, the mice were boosted with peptide in CFA. The frequency of tetramer+ CD4 T cells that bound tetramer with high intensity was assessed as described in the legend to Fig. 2 C. (C) After 18 wk, the mice were boosted with peptide in CFA. Tetramer+ cells were measured as described in the legend to Fig. 1. Each point represents one mouse, and the line shows the mean of each group. This experiment was performed twice with similar results.
